Anti-Sorting Nexin-11 Antibody is an antibody against Sorting Nexin-11 for use in Western Blotting, Immunoprecipitation.
Western Blotting Analysis: 3.75 µg/mL from a representative lot detected sorting nexin-11 in 10 µg of HEK293T cell lysate.
Western Blotting Analysis: A representative lot detected reducing amount of sorting nexin-11 in lysates from HEK293 cells that had been transfected with SNX11 shRNA (Joyal, J.S., et al. (2014). Nat. Med. 20(10):1165-1173).
Western Blotting Analysis: A representative lot detected the presence of sorting nexin-11 in F2RL1-GFP fusion affinity pull-down only when using lysates of F2RL1-GFP-transfected HEK293 cells that had been stimulated with the F2RL1 agonist peptide (F2RL1-AP) SLIGKV, but not unstimulated F2RL1-GFP transfectants (Joyal, J.S., et al. (2014). Nat. Med. 20(10):1165-1173).
Immunoprecipitation Analysis: A representative lot co-immunoprecipitated importin-β1 (Impβ1) and F2RL1 with sorting nexin-11 from lysates of HEK293 cells that had been stimulated with the F2RL1 agonist peptide (F2RL1-AP) SLIGKV (Joyal, J.S., et al. (2014). Nat. Med. 20(10):1165-1173).

General description
Sorting nexin-11 (UniProt Q91WL6) is encoded by the Snx11 gene (Gene ID 74479) in murine species. Sorting nexins (SNXs) are involved in diverse intracellular endosomal traffickings. Their phox homology (PX) domain binds certain phosphatidylinositols (PIs) and mediates their recruitment to PI-rich vesicles. Most SNXs contain additional recognizable domains and are referred to as SNX PX-BAR or SNX PX-other, while SNX3, SNX10, SNX11, SNX12, SNX22, and SNX24 contain only the PX domain and are referred to as SNX PX. SNX11 is found to contain a novel PXe domain (a.a. 16-132 of human and mouse SNX11) with two additional α-helices next to the conventional PX domain structure composed of three β-strands and three α-helices. F2RL1 (a.k.a. PAR2) is a protease-activated GPCR highly expressed throughout the central and the peripheral nervous system. F2RL1 is reported to translocate from the plasma membrane to nucleus upon stimulation via a microtubule-dependent shuttle machinery that requires sorting nexin 11.

This polyclonal antibody reacts with both human and murine sorting nexin-11. The immunogen sequence is also present in human spliced isoform 2 and rat sorting nexin-11.
